چکیده

B Background The pathogenic bacterium Listeria monocytogenes is able to cross the endothelial, placental and blood-brain barrier causing gastritis, meningitis, neonatal death and other diseases in humans. PrfA (Positive regulatory factor A) is the sole regulator of the virulence genes identified in Listeria monocytogenes till date. It regulates genes within the virulence gene cluster (vgc), which harbors prfA itself, plcA, hly, mpl, actA and plcB. PrfA is also known to regulate cell-wall associated internalins (inlA and inlB), secreted internalins (inlC) and activates the transcription of genes involved in hexose phosphate uptake and bile salt hydrolases. We tried to identify additional PrfA regulated genes using a novel bioinformatic approach.

The Listeria monocytogenes σB Regulon and Its Virulence-Associated Functions Are Inhibited by a Small Molecule

UNLABELLED The stress-responsive alternative sigma factor σ(B) is conserved across diverse Gram-positive bacterial genera. In Listeria monocytogenes, σ(B) regulates transcription of >150 genes, including genes contributing to virulence and to bacterial survival under host-associated stress conditions, such as those encountered in the human gastrointestinal lumen. An inhibitor of L. monocytogene...

Microarray-based characterization of the Listeria monocytogenes cold regulon in log- and stationary-phase cells.

Whole-genome microarray experiments were performed to define the Listeria monocytogenes cold growth regulon and to identify genes differentially expressed during growth at 4 and 37 degrees C. Microarray analysis using a stringent cutoff (adjusted P < 0.001; >/=2.0-fold change) revealed 105 and 170 genes that showed higher transcript levels in logarithmic- and stationary-phase cells, respectivel...

Isolation of Listeria monocytogenes mutants with high-level in vitro expression of host cytosol-induced gene products.

The facultative intracellular bacterial pathogen Listeria monocytogenes dramatically increases the expression of several key virulence factors upon entry into the host cell cytosol. actA, the protein product of which is required for cell-to-cell spread of the bacterium, is expressed at low to undetectable levels in vitro and increases in expression more than 200-fold after L. monocytogenes esca...

Listeria monocytogenes B Modulates PrfA-Mediated Virulence Factor Expression †

Listeria monocytogenes B and positive regulatory factor A (PrfA) are pleiotropic transcriptional regulators that coregulate a subset of virulence genes. A positive regulatory role for B in prfA transcription has been well established; therefore, observations of increased virulence gene expression and hemolytic activity in a sigB strain initially appeared paradoxical. To test the hypothesis that...

Biosynthesis of Silver Nanoparticles Using Chlamydomonas reinhardtii and its Inhibitory Effect on Growth and Virulence of Listeria monocytogenes

Background: Biosynthesis of nanoparticles using microorganisms, enzymes, and plant extracts is regarded as an alternative to chemical methods. Microalgae appear to be an efficient biological platform for nanoparticle synthesis as they grow rapidly and produce large biomass at lower cost. Objectives: The possibility of silver nanoparticles biosynthesisby freshwater green microalgae, Chlamydomona...
